Les personnes souffrant d'aniridie sont habituellement atteintes de basse vision d\u00e8s la naissance et d\u00e9veloppent \u00e9ventuellement un glaucome et des cataractes, lesquels peuvent mener \u00e0 la c\u00e9cit\u00e9. La plupart des cas sont dus \u00e0 une mutation du g\u00e8ne PAX6. Bien que les traitements actuels puissent ralentir la progression de la maladie, il n'existe aucun rem\u00e8de ni traitement contre cette affection.<\/p>\n\n\n\n<p>Gr\u00e2ce au financement de Vaincre la c\u00e9cit\u00e9 Canada, l'\u00e9quipe de la D<sup>re<\/sup> Simpson a mis au point une th\u00e9rapie g\u00e9nique \u00e0 effraction minimale pour traiter l'aniridie et elle la teste pr\u00e9sentement sur un mod\u00e8le animal.<\/p>\n\n\n\n<h5 class=\"wp-block-heading\">Des th\u00e9rapies g\u00e9niques pour traiter les maladies oculaires<\/h5>\n\n\n\n<p>La plupart des th\u00e9rapies g\u00e9niques actuelles pour traiter les maladies oculaires h\u00e9r\u00e9ditaires introduisent le nouveau g\u00e8ne directement dans l'\u009cil, la plupart du temps avec une injection sous-r\u00e9tinienne, un processus d\u00e9licat. L'injection de la th\u00e9rapie g\u00e9nique dans l'\u009cil garantit que les cellules cibles re\u00e7oivent une dose suffisante tout en r\u00e9duisant la quantit\u00e9 de th\u00e9rapie g\u00e9nique qui atteint d'autres parties du corps, dans l'espoir de minimiser le risque d'effets secondaires.<\/p>\n\n\n\n<p>La D<sup>re<\/sup> Simpson effectue pr\u00e9sentement des tests pour savoir si la th\u00e9rapie g\u00e9nique pour les maladies oculaires pourrait \u00eatre administr\u00e9e sans injection dans l'\u009cil.<\/p>\n\n\n\n<figure class=\"wp-block-image size-full is-resized\"><img fetchpriority=\"high\" decoding=\"async\" src=\"https:\/\/www.optiknow.ca\/wp-content\/uploads\/2023\/09\/Dr.-Elizabeth-Simpson.png\" alt=\"\" class=\"wp-image-102500\" style=\"width:274px;height:274px\" width=\"274\" height=\"274\"\/><figcaption class=\"wp-element-caption\"> Dre Elizabeth Simpson<\/figcaption><\/figure>\n\n\n\n<p>L'\u00e9quipe de la D<sup>re<\/sup> Simpson a test\u00e9 la th\u00e9rapie g\u00e9nique par injection intraveineuse; la th\u00e9rapie \u00e9tant inject\u00e9e directement dans la circulation sanguine. Non seulement cette m\u00e9thode est moins invasive qu'une injection sous-r\u00e9tienne, mais parce que les mutations du g\u00e8ne PAX6 peuvent avoir un impact sur d'autres organes, elle pourrait aussi att\u00e9nuer les sympt\u00f4mes dans plusieurs parties du corps, y compris l'\u009cil.<\/p>\n\n\n\n<p>Dans le cadre de la bourse de recherche, l'\u00e9quipe de la D<sup>re<\/sup> Simpson a produit et test\u00e9 de multiples th\u00e9rapies g\u00e9niques PAX6 dans un mod\u00e8le animal afin d'identifier l'approche optimale.<\/p>\n\n\n\n<p>Apr\u00e8s avoir test\u00e9 diff\u00e9rentes doses dans des \u00e9tudes sur les animaux, la D<sup>re<\/sup> Simpson \u00e9tait ravie de d\u00e9montrer que la th\u00e9rapie g\u00e9nique PAX6 administr\u00e9e par injection intraveineuse avait atteint les cellules r\u00e9tiniennes et qu'elle \u00e9tait toujours pr\u00e9sente dans les cellules cinq mois apr\u00e8s l'injection. Fait important, il n'y avait aucune \u00ab toxicit\u00e9 \u00bb provenant du traitement.<\/p>\n\n\n\n<p>Suite \u00e0 l'administration de la th\u00e9rapie g\u00e9nique, l'\u00e9quipe n'a constat\u00e9 aucun grand impact sur la vision et elle pense qu'un plus grand nombre de cellules devront recevoir la th\u00e9rapie pour qu'une am\u00e9lioration se produise.
